Abstract CONTEXT: Commonly used immunoassays are not free from interference, which can be a confounder in the interpretation of test results. We present a case with extremely high multiple hormone levels due to such interference. CASE DESCRIPTION: A 33-year-old woman with no specific symptoms had markedly elevated TSH with normal free T4 and free T3 levels. Repeated measurements revealed discordantly high TSH, ACTH, FSH, PTH, IGF-1, prolactin, beta-human chorionic gonadotropin, and calcitonin levels without the associated clinical pictures. The measurements were repeated with the same patient sample on four different analytical platforms using chemiluminescence immunoassays/electrochemiluminescence immunoassays, and the results were divergent on each platform. Serial dilutions of serum samples revealed nonlinearity, suggesting assay interference. All hormonal measurements were in the normal range when heterophile antibody blocking tubes were used. The serum of the patient was then subjected to polyethylene glycol precipitation. The post-polyethylene glycol recovery resulted in hormone levels in the normal range. The patient did not receive any medications and has been under follow-up without any signs and symptoms for 24 months. CONCLUSIONS: This report illustrates a rare case of falsely elevated hormone levels due to assay interference caused by heterophile antibodies. We point out the importance of a close collaboration between clinicians and the laboratory to avoid unnecessary clinical investigations as well as inappropriate treatments.

Abstract The literature on entrepreneurship bears a distinctly positive cast, often with good reason. Entrepreneurs and their innovations have contributed enormously to national wealth, and so scholars have examined the personalities, capabilities, and contexts underlying these contributions. However, despite some early work, the negative aspects of the entrepreneurial personality have been largely ignored. We shall argue that given the nature of the challenges facing many entrepreneurs and the consequent demands of their jobs, certain personality traits will be quite valuable to them. These, however, tend to be Janus-faced in that positive attributes, such as energy, self-confidence, need for achievement, and independence, may sometimes devolve naturally into aggressiveness, narcissism, ruthlessness, and irresponsibility. Given the costly repercussions of the latter characteristics, we urge more study of their nature and causes among entrepreneurs.

Abstract BACKGROUND: Timely detection of graft rejection is an important issue in the follow-up care after solid organ transplantation. Until now, biopsy has been considered the “gold standard” in the diagnosis of graft rejection. However, non-invasive tests such as monitoring the levels of cell-free DNA (cfDNA) as a sensitive biomarker for graft integrity have attracted increasing interest. The rationale of this approach is that a rejected organ will lead to a significant release of donor-derived cfDNA, which can be detected in the serum of the transplant recipient. METHODS: We have developed a novel quantitative real-time PCR (qPCR) approach for detecting an increase of donor-derived cfDNA in the recipient’s serum. Common insertion/deletion (InDel) genetic polymorphisms, which differ between donor and recipient, are targeted in our qPCR assay. In contrast to some other strategies, no specific donor/recipient constellations such as certain gender combinations or human leukocyte antigen (HLA) discrepancies are required for the application of our test. RESULTS: The method was first validated with serial dilutions of serum mixtures obtained from healthy blood donors and then used to determine donor-derived cfDNA levels in patients’ sera within the first 3 days after their kidney transplantation had been performed. CONCLUSIONS: Our method represents a universally applicable, simple and cost-effective tool which can potentially be used to detect graft dysfunction in transplant recipients.

Abstract The communication strategy of most crickets and bushcrickets typically consists of males broadcasting loud acoustic calling songs, while females perform phonotaxis, moving towards the source of the call. Males of the pseudophylline bushcricket species Onomarchus uninotatus produce an unusually low-pitched call, and we found that the immediate and most robust response of females to the male acoustic call was a bodily vibration, or tremulation, following each syllable of the call. We hypothesized that these bodily oscillations might send out a vibrational signal along the substrate on which the female stands, which males could use to localize her position. We quantified these vibrational signals using a laser vibrometer and found a clear phase relationship of alternation between the chirps of the male acoustic call and the female vibrational response. This system therefore constitutes a novel multimodal duet with a reliable temporal structure. We also found that males could localize the source of vibration but only if both the acoustic and vibratory components of the duet were played back. This unique multimodal duetting system may have evolved in response to higher levels of bat predation on searching bushcricket females than calling males, shifting part of the risk associated with partner localization onto the male. This is the first known example of bushcricket female tremulation in response to a long-range male acoustic signal and the first known example of a multimodal duet among animals.

Abstract Systematic investigations and phylogenetic analyses have indicated that Astelia, as currently circumscribed, is paraphyletic, with Collospermum nested within it. Further, A. subgenus Astelia is polyphyletic, and A. subgenera Asteliopsis and Tricella are paraphyletic, as currently circumscribed. Revision of the subgeneric classification of Astelia is warranted to ensure classification accurately reflects the evolutionary history of these taxa. Collospermum is relegated to synonymy within Astelia. Astelia is dioecious or polygamodioecious, with a superior ovary, anthers dorsi- or basifixed, pistillodes or pistils that have a single short or poorly defined style, a 3 lobed stigma, and fleshy uni- or trilocular fruit with funicular hairs that are poorly to well developed. Astelia subgenus Collospermum (Skottsb.) Birch is described. A key to Astelia sections is provided. Astelia hastata Colenso, A. montana Seem., and A. microsperma Colenso pro parte are resurrected and the new combination A. samoense (Skottsb.) Birch, comb. nov. is made.

Abstract We investigate the population of cool, evolved stars in the Local Group dwarf elliptical galaxy M32, using Infrared Array Camera observations from the Spitzer Space Telescope. We construct deep mid-infrared colour-magnitude diagrams for the resolved stellar populations within 3.5 arcmin of M32's centre, and identify those stars that exhibit infrared excess. Our data are dominated by a population of luminous, dust-producing stars on the asymptotic giant branch (AGB) and extend to approximately 3 mag below the AGB tip. We detect for the first time a sizeable population of `extreme' AGB stars, highly enshrouded by circumstellar dust and likely completely obscured at optical wavelengths. The total dust-injection rate from the extreme AGB candidates is measured to be 7.5 × 10-7 M⊙ yr-1, corresponding to a gas mass-loss rate of 1.5 × 10-4 M⊙ yr-1. These extreme stars may be indicative of an extended star formation epoch between 0.2 and 5 Gyr ago.
